Nettet18. jan. 2024 · In this study we present a protocol for intact nuclei isolation from diatoms that was successfully applied to three different species: two pennates, Pseudo-nitzschia multistriata and Phaeodactylum tricornutum, and one centric diatom species, Chaetoceros diadema. NettetOur strategy was to transgenically tag nuclei in specific cell types and then use affinity isolation to purify them from the total pool of nuclei derived from a tissue, thereby avoiding the need for dis-sociating and mechanically purifying whole cells. We refer to this strategy as INTACT, for ‘isolation of nuclei tagged in specific cell types’.
